卵巢癌患者外周血及腹水癌细胞多耐药基因及凋亡因子检测

  摘 要:目的 探讨卵巢癌细胞多耐药基因表达和化疗药物促使卵巢癌细胞凋亡的作用。方法 从62例首次手术及17例复发卵巢癌患者腹水中提取富含癌细胞的悬液,应用3种荧光单克隆抗体P170、CD95及Apo2.7分别标记,经流式细胞仪检测。结果 ①化疗后癌细胞CD95及Apo2.7含量均高于化疗前(P<0.05);②腹腔化疗前后P170含量无明显差异;③复发组癌细胞P170含量显著高于初发手术组(P<0.05),且本组化疗后癌细胞CD95和Apo2.7含量明显低于初发手术组(P<0.05);④化疗可使外周血淋巴细胞CD95含量显著增加(P<0.05)。结论 腹腔灌注化疗药物对卵巢腹水癌细胞凋亡有明显的促进作用,但癌细胞多耐药基因的表达可显著减少癌细胞凋亡。

Study of cellular multidrug resistant gene and apoptosis factor in peripheral blood and ovarian carcionoma cells in abdominal cavity infusion chemotherapy

  Abstract:Objective To explore the multidrug resistant gene (Mdrl) expreession(P170) and cellular apoptosis of ovarian carcinoma in abdominal cavity infusion chemotherapy.Methods The samples of ascites cancer cells(ACC) were selectively collected in sixty-two primary and seventeen recurred ovarian carcinomas,immunologically marked by three kinds of fluorescent monoantibodies-P170(p-gp),CD95(Fas-Apo-1),Apo2.7,then determined by flow cytometry.Results ①The CD95 and Apo2.7 contents of ACC after chemotherapy were significantly higher than that before treatment(P<0.05);②The contents of P170 after chemotherapy were similar to that before treatment (P<0.05);③The P170 contents in recurred group were significantly higher than that found in the primary cancer group(P<0.05),and the contents of CD95 and Apo2.7 in the recurred group after chemotherapy were lower than that in the primary group.④The CD95 contents of peripheral lymphocyte were also higher after abdominal cavity infusion chemotherapy(P<0.05).Conclusions The abdominal cavity infusion chemotherapy could obviously improve the apoptostic rate of ACC in ovarian carcinoma;but the apoptostic rate may decrease when the content of P170 becomes elevated.
